Genetic Contribution to Cerebral Palsy Uncovered in Canadian-led Study

A recent study led by Canadian researchers has shed light on the genetic factors that may play a role in the development of cerebral palsy (CP). CP is a prevalent childhood-onset physical disability that affects motor skill development in children. While the condition has long been attributed to environmental factors such as infections, injuries, or lack of oxygen during birth, the genetic contributors to CP have remained largely unknown. However, the groundbreaking research conducted by scientists from The Hospital for Sick Children, the Research Institute of the McGill University Health Centre, and Holland Bloorview Kids Rehabilitation Hospital has provided new insights into the genetic underpinnings of CP.

Published in Nature Genetics, the study involved whole-genome sequencing of 327 children with CP, along with their biological parents. The researchers compared the genetic data to various clinical and pediatric control cohorts to identify potential genetic variants associated with CP. The findings revealed that over 11% of the children had a genetic variant or likely genetic variant linked to their CP, while nearly 18% had variants of uncertain significance that may be correlated with the condition. Moreover, many of these variants overlapped with other neurodevelopmental disorders, such as autism spectrum disorder (ASD), highlighting the complexity and diversity of CP and its genetic causes.

The discovery of these genetic variants in children with CP has significant implications for the diagnosis and treatment of the condition. By incorporating precision medicine programs and genetic testing into clinical practice, healthcare professionals can better understand the individualized risk factors associated with CP, allowing for tailored treatment approaches. Dr. Maryam Oskoui, one of the study co-leads, emphasized the importance of genetic testing in all children with CP to personalize future interventions and improve patient care.

Advancing Precision Health Programs

The data collected from this study have been made accessible through the Brain-CODE analytics and informatics platform, managed by the Ontario Brain Institute, with the aim of facilitating global access to genome sequencing data. This initiative not only promotes collaboration among scientists but also paves the way for the development of targeted treatment options for children with CP. Dr. Darcy Fehlings, a co-first author of the study, emphasized the significance of precision medicine in accurately diagnosing the etiology of CP and optimizing interventions for affected children.
